The large majority of chronic pain cases are considered to be neuropathic in origin, but effective treatment for neuropathic pain has been elusive. The goal of the current project is to evaluate a novel therapeutic approach that aims to reduce neuropathic pain by targeting pain-related neural plasticity at the neuromuscular junction. Recently, we have demonstrated that the enzyme tyrosine kinase A (TrkA) is a novel target for the treatment of neuropathic pain. We have also shown that TrkA is expressed at the neuromuscular junction, and that TrkA blockade with the selective inhibitor, K252a, reduces nerve injury-induced neuropathic pain. Importantly, this effect appears to involve reduction of central sensitization. Here, we propose to examine the role of TrkA activation at the neuromuscular junction in the generation of neuropathic pain using the model of chronic constriction injury (CCI) of the rat tibial nerve. Thus, we aim to: 1) define the contribution of TrkA signaling at the neuromuscular junction in the generation of neuropathic pain; 2) determine the contribution of TrkA signaling in the dorsal root ganglion in the generation of neuropathic pain; and 3) develop an in vitro system to test the role of TrkA signaling at the neuromuscular junction in the generation of neuropathic pain. This proposed research is innovative because it will provide a better understanding of how the neuromuscular junction contributes to the generation of neuropathic pain, and may lead to the development of a new class of pain therapies. The success of this project may open up a new approach to the treatment of neuropathic pain.Between the 6th and 8th August (2017), we will be hosting the first FEXPO for the FEX (Fédération des Entreprises Françaises) in Germany.
